Your resume will get you the interview, but being able to confidently answer questions in an interview will get you the offer. You need to have a good knowledge of data structures, simple algorithms, etc.. It doesn't matter whether you know asp.net or every function in the php library if you don't know how to reverse a linked list and get all the permutations of a string.
